It was built around a place of worship, of Tartessus origins, its foundations were tightly linked to the name of Luci Feri Fanum -sanctuary of the Lucero (Bright Star) or of the Crepúsculo (Twilight). Its first Christian name was Sant Lucar or Sant Lugar, redounding to the sacred origins and others attribute it to the Santo Evangelista (Holy Evangelist), whose commemoration was to coincide with the reconquest of the city. Sanlúcar was deinitively reconquered by Alfonso X in 1264. Sancho IV gave it up to the feudal estate of Alfonso Pérez de Gúzman in 1295.

The discovery of America turned Sanlúcar into a great trading port, from where important expeditions left: Colombus on his third visit in 1498, as well as Magallanes and Juan Sebastián Elcano to take part in their first trip around the world.
